概述

本課程將從基礎分子生物學與遺傳工程階段入門,介紹基因如何經由轉錄轉譯而產生蛋白質以及常用之基因轉殖與基因剔除小鼠的原理(三小時)。在了解蛋白質如何生成之後,將進一步介紹蛋白質的結構與相關偵測技術如西方墨點法與免疫螢光染色法等(三小時)。接著針對動物細胞的基本結構與組成,包括細胞核、細胞膜、細胞質以及包含於其中之胞器等作一概略性的介紹,並進一步介紹神經系統細胞的種類(如各種不同的神經元與膠質細胞等)與其主要功能,比較神經系統細胞與其他系統細胞相同與相異之處(三小時),之後再介紹顯微鏡的基本原理(三小時)。除了結構性的介紹神經細胞之外,本課程也將對神經元的功能性作一簡介,利如動作電位的產生原理與其生理意義等,屬於神經生理學的範疇(三小時)。最後介紹精神性疾病的動物行為模式(三小時)。總之,我們從不同層次提供完全不具備基本生物學知識的腦科所新生一個認識實驗神經科學的課程,以奠定其未來修讀進階腦科學相關課程的基礎。This course is composed of lectures of 6 series, 3 hours each. The first lecture will focus on the basic concepts of molecular biology and genetic engineering, including a brief introduction to RNA transcription, protein translation, as well as construction of transgenic mice and knockout mice. Introduction to protein science, including basic structural biology and commonly used experimental methods for detection of target proteins such as Western blotting and immunohistochemistry, will be included in the 2nd lecture. Following the basic concepts of molecular biology and commonly used experimental methods in molecular/cellular biology is the introduction of basic cell biology, including nuclei, plasma membrane, cytosol and intracellular organelles therein. This lecture will also include the introduction of specific cell types in the nervous system. This is then followed by principles of microscopy and introduction to neurophysiology including the underlying mechanisms responsible for generation of action potential will be included in the fifth series of lectures. Finally, animal behavior model, particularly those for the psychiatric disorders, will be introduced in the last lecture. In summary, this course is designed for those students without basic knowledge in biology and neuroscience such that they can be equipped with essential capability to take more advanced courses in the fields of brain science and neuroscience in the following semester.

本課程專為腦科學研究所「臨床醫學組」與「神經資訊造影組」學生而開設,目標在於針對缺乏實驗神經科學相關知識的學生提供一概論性課程,為其後續修讀較深入之必修課程如「腦科學概論」奠定基礎。課程內容包括與神經系統相關之基礎分子細胞生物學(含顯微鏡原理)、神經生理學以及動物行為學等等。This is a basic course for those graduate students without prior background knowledge in biology or experimental neuroscience, such as the students from the “Clinical Medicine” division and “NeuroInformatics and NeuroImage” division in the Institute of Brain Science. The main objective of this course, therefore, is to equip these students with essential knowledge in molecular biology, cell biology, molecular neuroscience, neurophysiology and animal behavior such that they can handle more advanced courses like “Introduction to Brain Science” later in this graduate program.
