概述

Class Description: “Microbial Genetic Engineering " is a comprehensive, hands-on course designed to guide students through the fundamental techniques employed in the rapidly evolving fields of Genetic Engineering. The course covers a range of topics including plasmid construction, gene expression manipulation, bacterial host genomic modifications, enzyme bioprospecting, enzyme assay and pathway assay, protein engineering and direct evolution, and an introduction to the latest sequencing technologies. Through real-world examples, students will learn how to design, conduct, and interpret experimental results, and gain a deeper understanding of the practical aspects of these technologies. Purpose of the Class: The purpose of this course is to equip students with the practical skills and knowledge in Genetic Engineering, which are essential for contributing to research and development in the fields of Metabolic Engineering and Synthetic Biology. It aims to give students a solid foundation of key techniques, encourage critical thinking, promote innovative experimental design, and foster an understanding of the practical applications of these concepts in various applications, including biotechnology, biofuel and biochemical productions, and pharmaceuticals.

第 11 週Case study: bacterial host genomic modifications – gene deletion (with Assignments) (Using ptsG deletion for lambda-Red system, lsrF deletion for CRISPR system)
第 12 週Case study: Bacterial host genomic modifications – gene integration (using tal integration CRISPR and Transposon-encoded CRISPR–Cas systems)
第 13 週Sanger sequence applications
第 14 週NGS & Nanopore applications
