近年來基因體學技術的進步,使得族群遺傳學與分子演化學的理論可以實際應用於醫學上很多重要的領域。目前許多國家已先後啟動了大樣本數的群體基因體解密計劃來尋找每族群基因變異與疾病的關聯與病因機轉。其中,台灣人體生物資料庫、台灣精準醫療計畫等也已如火如荼地進行,並計劃要建立萬人以上的基因資料庫。為配合未來基因體生物學的研究的發展,本課程將介紹族群遺傳學與分子演化學的基礎理論並從演化學的觀點來探討人類的演化與疾病的關係,如何將族群遺傳學的理論應用於精準醫學的發展。本課程主要修課對象為主修生物學及醫學相關學系的大學部與研究所學生。 Recent advances in genomic technology has allowed application of theories in molecular population genetics and evolution to a wide range of disciplines for studying biological and medical sciences. In particular, the promise of precision medicine has led to the development of population-based human genome projects (usually >100,000 individuals) in many countries in a high hope of making a complete list of genetic variants underlying disease susceptibility. Such a gigantic dataset also provides us a great opportunity to study patterns of genetic variation and identify causal variants that contribute to disease susceptibilities. This course will introduce theories and analytic methods in molecular evolution with a special focus on studying evolutionary changes in allele frequency of genetic variants in a population (microevolution). The goal of this course is to provide basic knowledge for studying population genomics in biomedical sciences. Both undergraduate and graduate students are welcome to take this course although it is recommended to take an elementary statistics courses in advance before taking this course. PART I – Introduction to molecular population genetics and evolution (中文) 將介紹分子族群遺傳學、分子演化學的理論基礎。課程內容包含 (ㄧ)分子族群遺傳學的理論基礎:基因突變(mutation)、基因漂移(genetic drift)、合組理論(coalescence theory)、自然選擇 (natural selection), 族群遺傳結構(population structure)。(二)基因體族群資料分析:族群遺傳結構分析(population structure analysis)、遺傳連鎖不平衡分析 (LD-linkage disequilibrium analysis)、自然選擇印痕分析(selection signature detection analysis)。 PART II – evolutionary medicine 將從分子族群遺傳學、演化學的理論來分析許多疾病與人類演化歷史的關係。包括自然選擇與疾病的關係、人類與傳染病共演化關係、Covid 19的遺傳起源與演化、癌細胞的演化學理論、從人類演化歷史透視精準醫學的發展。 PART I – Introduction to molecular population genetics and evolution (English) This course will introduce theories in molecular population genetics and evolution. The course topics include: (1) fundamental theory in molecular population genetics: mutation, genetic drift, coalescence theory, natural selection, and genetic structure. (2) genomic data analysis: population structure analysis, LD- based analysis, and selection signature detection analysis. PART II – evolutionary medicine This course will also cover several contemporary topics in evolutionary molecular medicine including: the role of natural selection in disease susceptibility, co-evolution between human and infectious diseases, genetic origin and evolution of SARS-CoV-2, and theories in cancer-cell evolution. Finally, we will also provide some insights into the future development of precision medicine from an evolutionary perspective. All lectures will be delivered in English.
Completion of biostatistics and genetics courses in advance is recommended but not mandatory.
